產(chǎn)品介紹

DiR是一個(gè)親脂性、近紅外熒光花青染料。這個(gè)染料常用于標(biāo)記細(xì)胞質(zhì)膜。DiR的兩個(gè)18-碳鏈插入到細(xì)胞膜,從而進(jìn)行特定的、穩(wěn)定的細(xì)胞染色,幾乎不會(huì)發(fā)生細(xì)胞間的染料轉(zhuǎn)移。
DiR(近紅外熒光)和其他細(xì)胞膜熒光染料如DiI(橙色熒光),DiO(綠色熒光),DiD(紅色熒光)配合使用,為多色成像和流式細(xì)胞分析提供了有效的工具。
DiR染色后可進(jìn)行多聚甲醛(不可使用甲醇等其他試劑)的固定,但不建議在染色后進(jìn)行透化的過(guò)程。此外,在固定透化(室溫下用0.1% TritonX-100透化)后,也可以很好地進(jìn)行質(zhì)膜染色。
以每次使用100 μL染色工作液,染色工作液濃度5 μM計(jì)算,5 mg配置為工作液大概可以用9867次。

注意事項(xiàng)
1. 使用前請(qǐng)將產(chǎn)品瞬時(shí)離心至管底,再進(jìn)行后續(xù)實(shí)驗(yàn)。
2. DiR染色固定的細(xì)胞或組織樣品時(shí),通常使用配制在PBS中的4%多聚甲醛進(jìn)行固定,使用其它不適當(dāng)?shù)墓潭ㄒ簳?huì)導(dǎo)致熒光背景較高。
3. 熒光染料均存在淬滅問(wèn)題,請(qǐng)盡量注意避光,以減緩熒光淬滅。

細(xì)胞追蹤有很多產(chǎn)品,該如何選擇?
首先確定追蹤細(xì)胞的時(shí)間,然后考慮染料結(jié)合機(jī)制。
1.鈣黃綠素染料(C4041)標(biāo)記均勻且對(duì)短期細(xì)胞遷移示蹤效果極佳,但也會(huì)被某些類型細(xì)胞迅速外排。
2.親脂性的花青素染料,如DiI(D4010),DIO(D4007),DiA(D4059),DiD(D4019),DiR(D4006)能標(biāo)記細(xì)胞膜而不破壞其功能,并且能持續(xù)更長(zhǎng)時(shí)間,但如果發(fā)生膜融合則可能會(huì)染上其他細(xì)胞。此外,它們還會(huì)在透化過(guò)程中丟失。
3.CellTracker染料(C4060)更有利于長(zhǎng)期標(biāo)記,其帶有溫和的氯甲基反應(yīng)基團(tuán)使之能夠與細(xì)胞組分共價(jià)結(jié)合。
4.CFDA SE(C4070)也能共價(jià)地結(jié)合于細(xì)胞組分。
